Target Information
ICAD (Inhibitor of Caspase-3-Activated DNase) is a substrate that regulates nuclear apoptosis. It exists in two isoforms:
- ICAD-L/DFF45 (45 kDa) — functional isoform
- ICAD-S/DFF35 (35 kDa) — non-functional isoform
ICAD inhibits CAD (Caspase-Activated DNase) under normal conditions. During apoptosis, caspase-3 cleaves ICAD at residues 117 and 224, releasing CAD to induce internucleosomal DNA fragmentation. ICAD also functions as a CAD-specific folding chaperone. It has been studied as a potential target for restoring normal apoptotic signaling in colon and brain cancer cells. Two alternatively spliced transcript variants encoding distinct isoforms have been identified for the ICAD gene.
