Target Information
Tuberous sclerosis complex (TSC)는 TSC1 또는 TSC2 종양 억제 유전자의 변이에 의해 발생하는 상염색체 우성 종양 증후군입니다. 이들 유전자의 산물은 단백질 복합체를 형성하여 mammalian Target of Rapamycin (TOR) 신호를 간접적으로 감소시킵니다. TOR은 영양 수준과 성장 인자로부터 신호를 통합하여 세포 성장과 세포 주기를 조절하는 진화적으로 보존된 serine/threonine kinase입니다.
Rheb은 Ras superfamily에 속하는 GTP 결합 단백질로, GTP/GDP 비율이 높을 때 TOR 활성을 자극합니다. 면역침전된 TSC1/TSC2는 in vitro에서 Rheb GTPase 활성을 자극하는 것으로 나타나, TSC1/TSC2가 Rheb의 TOR 활성화 능력을 감소시킴을 시사합니다. 또한 TSC1과 TSC2의 과발현은 Rheb에 결합된 GTP/GDP 비율의 유의한 감소 및 세포 성장 억제를 초래합니다.
40 kDa 크기의 짧은 TSC1 isoform이 존재하지만 그 기능은 알려져 있지 않습니다.
