Target Information

PDZ-GEF1 functions as a guanine nucleotide exchange factor (GEF) that activates Rap and Ras family small GTPases by exchanging bound GDP for GTP in a cAMP-dependent manner. It serves as a link between cell surface receptors and intracellular signaling cascades involving Rap/Ras. Acts as an effector for Rap1 by direct association with Rap1-GTP, amplifying Rap1-mediated signaling. Exhibits weak activity on HRAS. Its ability to bind cAMP and cGMP remains controversial. Interaction with ligand-activated β1-adrenergic receptor (ADRB1) leads to Ras activation through the Gsα signaling pathway. Involved in cAMP-induced Ras and Erk1/2 signaling that inhibits long-term melanogenesis by reducing dendrite extension and melanin synthesis. Provides inhibitory signals for melanoma cell proliferation and promotes apoptosis independently of cAMP. Regulates cAMP-induced neuritogenesis via the Rap1/B-Raf/ERK pathway, independent of PKA and RAPGEF3/4. Plays roles in neuron migration, forebrain fiber connection formation, NGF-induced Rap1 activation, BDNF-induced axon outgrowth, embryonic blood vessel formation, and endothelial barrier function regulation.
