Background
Von Hippel-Lindau tumor suppressor (VHL) is associated with the dominantly inherited familial cancer syndrome known as Von Hippel-Lindau syndrome (VHL). Germline mutations in this gene cause susceptibility to various malignant and benign tumors. The VHL protein forms a complex with elongin B, elongin C, and cullin-2, exhibiting ubiquitin ligase E3 activity. It mediates ubiquitination and degradation of hypoxia-inducible factor (HIF), a key transcription factor regulating oxygen-dependent gene expression. RNA polymerase II subunit POLR2G/RPB7 is also a reported target. Alternatively spliced transcript variants encoding distinct isoforms have been identified.
