Background
This gene encodes the nuclear hormone receptor for vitamin D3, which also acts as a receptor for the secondary bile acid lithocholic acid. The receptor is part of the nuclear transcriptional regulatory factor family, showing similarity to steroid and thyroid hormone receptors. Its downstream targets are mainly involved in mineral metabolism but also regulate immune response and cancer-related pathways. Mutations in this gene are linked to type II vitamin D-resistant rickets. A single nucleotide polymorphism in the initiation codon can lead to an alternative translation start site, and alternative splicing results in multiple transcript variants encoding different proteins.
