Cellular Localization

Cytoplasm, cytosol, endoplasmic reticulum, nucleus, and stress granules. Present in neuronal inclusion bodies found in motor neurons from amyotrophic lateral sclerosis patients and Lewy bodies in Parkinson disease. Recruited to the cytoplasmic surface of the endoplasmic reticulum via interaction with AMFR/gp78. Also recruited to DNA double-strand break sites, stalled replication forks, and damaged lysosomes decorated with K48-linked ubiquitin chains. Colocalizes with TIA1, ZFAND1, and G3BP1 in cytoplasmic stress granules.

Background

Valosin-containing protein (VCP) in Homo sapiens is a member of a family of ATP-binding proteins involved in vesicle transport, 26S proteasome function, and peroxisome assembly. This structural protein associates with clathrin and heat-shock protein Hsc70 to form complexes. It participates in cellular events regulated during mitosis, including homotypic membrane fusion, spindle pole body function, and ubiquitin-dependent protein degradation.
