Background

This gene encodes a nuclear phosphoprotein that plays a crucial role in maintaining genomic stability and acts as a tumor suppressor. The encoded protein forms part of the BRCA1-associated genome surveillance complex (BASC), combining with other tumor suppressors, DNA damage sensors, and signal transducers. It associates with RNA polymerase II and interacts with histone deacetylase complexes through its C-terminal domain, contributing to transcription regulation, DNA double-strand break repair, and recombination. Mutations in this gene account for approximately 40% of inherited breast cancers and over 80% of inherited breast and ovarian cancers. Alternative splicing affects subcellular localization and function, generating multiple transcript variants.
