Background
Tryptases comprise a family of trypsin-like serine proteases belonging to the peptidase family S1. These enzymes are active only as heparin-stabilized tetramers and are resistant to all known endogenous proteinase inhibitors. Several tryptase genes are clustered on chromosome 16p13.3 and share distinct features such as highly conserved 3' UTRs and tandem repeat sequences that may regulate mRNA stability. These genes contain an intron immediately upstream of the initiator Met codon, separating transcription initiation from the protein coding sequence — a characteristic feature of tryptases. The alleles of this gene exhibit significant sequence variation, previously thought to represent two separate genes.
