Tryptases comprise a family of trypsin-like serine proteases, belonging to the peptidase family S1. These enzymes are active only as heparin-stabilized tetramers and are resistant to endogenous proteinase inhibitors. Multiple tryptase genes are located on chromosome 16p13.3 and share conserved 3′ UTR sequences with tandem repeats that may regulate mRNA stability. Each gene includes an intron upstream of the start codon, a feature typical of tryptases but rare in other genes. The alleles of this gene display high sequence variability, once thought to represent two distinct genes.
