Target Information

Mitochondrial transcription factor A (TFAM) is a nuclear-encoded protein that translocates to mitochondria, where it maintains and regulates mitochondrial DNA (mtDNA) replication and transcription. TFAM is essential for mitochondrial genome packaging, stability, and expression. It contains an LC3 interacting region (LIR) motif, enabling interaction with autophagy proteins such as LC3, crucial for the autolysosomal pathway that removes leaked mtDNA and reduces inflammation. TFAM levels vary by tissue and influence mitochondrial function and oxidative phosphorylation (OXPHOS). Dysregulation of TFAM can lead to mitochondrial dysfunction and diseases associated with mtDNA stress, including neurodegeneration.
