Product Specific Information

The XMG1.2 antibody reacts with mouse interferon (IFN) gamma and is a neutralizing antibody.
Mouse IFN gamma (~20 kDa) is produced by activated T, B, and NK cells, acting as an antiviral and antiparasitic cytokine. It inhibits proliferation of normal and transformed cells in synergy with cytokines such as TNF alpha.

  • Applications Reported: Intracellular staining followed by flow cytometric analysis.
  • Applications Tested: Intracellular staining of mouse splenocytes using the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
    • Recommended ≤1.0 µg/test (100 µL volume, 10⁵–10⁸ cells/test).
    • Optimize antibody titration for best performance.

Brilliant Violet™ 605 (BV605) emits at 605 nm and is suitable for cytometers with a 405 nm violet laser.
When using multiple polymer dye-conjugated antibodies, use:

  • Super Bright Complete Staining Buffer (Product # SB-4401-42), or
  • Brilliant Stain Buffer™ (Product # 00-4409-75)

Light Sensitivity: Protect from light to avoid photo-induced oxidation.
Fixation: Compatible with IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C.
Not compatible with methanol-based fixation.

Target Information

IFN gamma (Interferon gamma, Type II interferon)

  • Produced mainly by T lymphocytes and NK cells in response to antigens and cytokines.
  • Dimeric glycoprotein (~45 kDa) composed of two 146-amino-acid subunits.
  • Functions: antiviral activity, tumor antiproliferative effects, induction of MHC I/II, macrophage activation, enhanced immunoglobulin secretion.
  • Highly species-specific; human IFN gamma does not cross-react with mouse IFN gamma.
  • Activation via IFN gamma receptor I/II and JAK-STAT pathway.
  • Upregulated by IL2, FGF basic, EGF; downregulated by vitamin D3 or DMN.
  • Mutations are associated with aplastic anemia.
