Target Information
MERTK (cMER) is a tyrosine kinase proto-oncogene involved in the retinal pigment epithelium (RPE) phagocytosis pathway, which is associated with retinal disease. It regulates cell survival, migration, differentiation, and efferocytosis. Ligand binding induces autophosphorylation of MERTK, creating docking sites for downstream signaling molecules such as GRB2 and PLCG2, leading to phosphorylation of MAPK1, MAPK2, FAK/PTK2, or RAC1. MERTK signaling is crucial for macrophage clearance of apoptotic cells, platelet aggregation, cytoskeleton reorganization, and inhibition of TLR-mediated innate immune responses via STAT1 activation, promoting SOCS1 and SOCS3 production.
