Target Information
STAT3는 STAT (signal transducers and activators of transcription) 단백질 패밀리에 속하며, 수용체 연관 키나아제에 의해 인산화되어 핵으로 이동 후 전사인자로 작용합니다. CREB-binding protein 등의 공동활성화 인자가 STAT3의 전사 활성화에 필요합니다.
STAT3는 Interferon-α, Interferon-γ, EGF, PDGF, IL6 등에 의해 활성화되며, Tyr705 위치의 인산화는 JAK1 및 JAK2에 의해 매개되어 이합체 형성, 핵 이동 및 DNA 결합에 필수적입니다.
STAT3 유전자는 인간 염색체 17q에 위치하며, IFN-α에 의해 활성화되지만 IFN-β에는 반응하지 않습니다. c-Jun 및 CREB와 같은 전사 인자와 연관되어 있으며, 세포 성장, 아폽토시스 등 다양한 세포 과정에 관여합니다.
Rac1 GTPase는 STAT3의 활성을 조절하며, PIAS3 단백질은 STAT3의 특이적 억제 인자로 알려져 있습니다. 세 가지 대체 스플라이싱 변이체가 보고되었으며, STAT3 유전자 결실은 생쥐 배아 단계에서 치명적입니다.
