Product Specific Information

Qdot™ Antibody conjugates exhibit bright fluorescence emission, suitable for detecting low-abundance extracellular proteins. Similar in size to R-phycoerythrin (R-PE) and compatible with organic fluorophore conjugates, they can be excited with any wavelength below emission maximum—optimally UV or violet light.

• Narrow, symmetric emission profiles minimize compensation in multicolor assays.
• Best used with 405 nm violet laser for efficient multicolor analysis.
• Compatible with standard staining buffers (PBS with 1% BSA).
• Analyze stained cells within 18 hours; use diluted reagent within one day.
• Compatible with Cal-Lyse™ and FIX & PERM™ reagents.
• Some BD FACS™ Lysing Solution batches may interfere with fluorescence.
• Tested by flow cytometry using human peripheral blood leukocytes.

Instrument Setup:

• Optimal excitation: UV or 405 nm light (also excitable at 488 nm).
• Ensure cytometer has appropriate emission filters (recommended: 605/20 nm).
• Check for spectral overlap when using multiple lasers.

Storage:

• Store at 2–8°C in the dark.
• Do not freeze.
• Shelf life: 6 months from receipt unless otherwise indicated.

Target Information

CD4 antigen recognizes MHC class II molecules and acts as a co-receptor for HIV. Expressed primarily in T helper cells, and also in monocytes, macrophages, and dendritic cells. Tissue-level expression found in thymus, lymph nodes, tonsils, spleen, and specific regions of brain and gut.

CD4 initiates or enhances early T-cell activation via association with the T-cell receptor complex and Lck kinase. It can mediate neuronal damage in infectious or immune-mediated CNS diseases. Multiple alternatively spliced transcripts have been identified (RefSeq, July 2017).
