Product Specific Information

The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and virtually all mature T cells and is upregulated upon T-cell stimulation. It binds to CD70 and plays an important role in T cell–B cell interaction.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on normal human peripheral blood cells. Recommended use is 5 µL (0.8 µg) per test in 100 µL total volume, with 10⁵–10⁸ cells/test.

Compatibility Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample before combining with cells.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily, primarily expressed on thymocytes, T cells, and a subset of B cells. It regulates lymphocyte activation, proliferation, and apoptosis through interaction with CD70. Soluble CD27 is a prognostic marker in B cell malignancies. CD27 dysfunction is associated with Lymphoproliferative Syndrome 2 and Autosomal Recessive Lymphoproliferative Syndrome.
