Target Information

The expression pattern of the Prox1 homeobox gene suggests that it plays a role in various embryonic tissues, including the lens. Prox1 mRNA is present in many human tissues, with the highest expression in the lens. Homozygous Prox1- mice die at midgestation due to developmental defects, particularly affecting lens development. Prox1 inactivation leads to abnormal cell proliferation, downregulation of Cdkn1b and Cdkn1c, misexpression of E-cadherin, and excessive apoptosis, resulting in defective lens formation.

Prox1 is expressed in endothelial cells that contribute to lymphatic system formation and acts as a key regulator of lymphatic development. It is also required for hepatocyte migration in mice; loss of Prox1 results in smaller livers with reduced hepatocyte clustering. In the embryonic mouse retina, Prox1 regulates progenitor cell cycle exit, influencing proliferation and cell fate determination.
