Target Information
Follistatin-like protein 1 (FSTL1) is a widely-expressed extracellular glycoprotein belonging to the osteonectin (BM-40/SPARC) family. It contains both a follistatin-like and extracellular calcium-binding domain. Initially identified as a TGF-beta-inducible protein in mouse osteoblasts, FSTL1 regulates proliferation, differentiation, apoptosis, and migration. It participates in processes such as embryonic development, inflammatory response, angiogenesis, tumorigenesis, and immune disease pathogenesis.
FSTL1 is highly conserved across mammals and widely expressed in human tissues. It can be upregulated by innate immune mediators like TLR4 agonists and IL-1β through NFκB pathways, inducing cytokines such as TNF-α, IL-1β, IL-6, and IL-8. While mesenchymal cells produce FSTL1, hematopoietic cells normally do not. Macrophages and monocytes can uptake FSTL1 at inflammation sites, leading to caspase-1 activation and IL-1β processing. Overexpression is linked to rheumatoid arthritis and osteoarthritis, while reduced serum levels are associated with ovarian and endometrial cancers.
