Target Information

Mitochondrial transcription factor A (TFAM) is a crucial protein encoded by nuclear genes but transported to mitochondria, where it maintains and expresses mitochondrial DNA (mtDNA). It regulates mtDNA replication and transcription, ensuring mitochondrial function. TFAM contains an LC3 interacting region (LIR) motif that binds autophagy proteins such as LC3, supporting the autolysosomal pathway to remove leaked mtDNA and limit inflammation.

TFAM levels vary across tissues, influencing mitochondrial function. Elevated levels can repress mtDNA expression in skeletal muscle, affecting oxidative phosphorylation (OXPHOS), while in cardiac tissue, balanced TFAM-to-mtDNA ratios preserve OXPHOS capacity. TFAM is essential for mitochondrial genome packaging, stability, and replication, and its dysregulation is linked to neurodegeneration and mtDNA stress-related diseases.
