Target Information
Insulin and Insulin-like Growth Factor receptors are transmembrane tyrosine kinase receptors that play critical roles in development, cell growth, and metabolism. Insulin/IGF-1 binding to the extracellular domain of IR and IGFR leads to autophosphorylation and activation of tyrosine kinase activity, enabling substrate phosphorylation. Unique phosphorylation sites within each receptor contribute to signaling specificity. The tyrosine kinase domains contain a three-tyrosine motif—Tyr1158, Tyr1162, Tyr1163 for IR and Tyr1131, Tyr1135, Tyr1136 for IGF1R. Autophosphorylation initiates at Tyr1162 (or Tyr1135) and proceeds sequentially, resulting in full activation of IR and IGF1R.
