Target Information

IRF3 (Interferon Regulatory Factor 3) is a key transcriptional regulator of type I interferon (IFN)-dependent immune responses. It plays a critical role in the innate immune response against DNA and RNA viruses.
IRF3 regulates the transcription of type I IFN genes (IFN-α and IFN-β) and IFN-stimulated genes (ISG) by binding to an interferon-stimulated response element (ISRE) in their promoters. It acts as a more potent activator of the IFN-β gene than the IFN-α gene and is essential for both early and late phases of IFNA/B gene induction.
In uninfected cells, IRF3 resides in the cytoplasm in an inactive form. Upon viral infection or toll-like receptor signaling, it is phosphorylated by IKBKE and TBK1 kinases, leading to dimerization, nuclear localization, and association with CREB-binding protein to form the dsRNA-activated factor 1 (DRAF1) complex. This complex activates transcription of type I IFN and ISG genes.
Mutations in the IRF3 gene can cause acute infection-induced herpes-specific encephalopathy type 7.
