Product Specific Information

This DS7HAXL monoclonal antibody recognizes human Axl, a member of the TAM family of tyrosine kinase receptors that includes MerTK and Tyro3.
It is suitable for flow cytometry on both native and paraformaldehyde-fixed cells, and also stains formalin-fixed, paraffin-embedded tissues for IHC analysis.

Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
Use at 5 µL (0.125 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Brilliant Violet™ 421 (BV421) emits at 423 nm and is used with a 405 nm violet laser.
Ensure instrument compatibility before use.

When using multiple polymer dye-conjugated antibodies (Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Target Information

AXL is a chronic myelogenous leukemia-associated oncogene also linked to colon cancer and melanoma.
It encodes a receptor tyrosine kinase that transduces extracellular signals into the cytoplasm via binding to growth factors such as GAS6.
TAM receptors (Tyro3, Axl, MerTK) are involved in apoptotic cell clearance. Mice lacking all TAM receptors develop autoimmune phenotypes similar to systemic lupus erythematosus.
Axl may also serve as an entry receptor for filoviruses and is used to identify Siglec 6+/Axl+ dendritic cells.
Expression can be upregulated by TLR ligands (LPS, poly I:C). Soluble Axl is generated by proteolytic cleavage, and elevated plasma levels may indicate inflammation or cancer.
