Antibody detects endogenous levels of total SHANK3.

Target Information

SHANK3 is a major scaffold postsynaptic density protein that interacts with multiple proteins and complexes to orchestrate dendritic spine and synapse formation, maturation, and maintenance. It connects receptors of the postsynaptic membrane, including NMDA-type and metabotropic glutamate receptors, via complexes with GKAP/PSD-95 and HOMER, and links to the actin-based cytoskeleton. SHANK3 plays a critical role in structural and functional organization of the dendritic spine and synaptic junction, interacting with Arp2/3 and WAVE1 complexes and promoting F-actin clustering. Through regulation of actin dynamics, SHANK3 contributes to neuron growth cone motility and NMDA receptor signaling, and modulates GRIA1 exocytosis and GRM5/MGLUR5 expression to control synaptic transmission and plasticity. It may be required at early stages of synapse formation and inhibited by IGF1 to promote synapse maturation. (Uniprot)
