Target Information
Transglutaminase (TGase)는 Ca²⁺ 결합을 필요로 하는 단백질 교차결합 효소로, e-(γ-glutamyl) lysine 이소펩타이드 결합을 형성합니다. 포유류에서는 8개의 TGase 동형효소가 확인되었으며, 그중 Tissue transglutaminase (tTGase, TGase 2)는 N-terminal β-sandwich, catalytic core, 두 개의 C-terminal β-barrel domain으로 구성됩니다.
tTGase는 세포 사멸, 증식, 분화 및 수용체 매개 내포작용에 관여할 수 있습니다. 알츠하이머병 뇌에서는 tTGase 활성 증가로 인해 Ab peptide, β-amyloid precursor protein, tau protein 등의 중합이 일어나며, 신경섬유 엉킴(neurofibrillary tangles) 형성에 기여할 수 있습니다.
