Target Information

Trpc6 forms a receptor-activated calcium channel in the cell membrane, activated by diacylglycerol (DAG) and regulated via the phosphatidylinositol second messenger system. Activation occurs independently of protein kinase C and intracellular calcium levels.
Defects in the TRPC6 gene are associated with focal segmental glomerulosclerosis 2 (FSGS2).
TRPC6 belongs to the mammalian transient receptor potential (TRP) superfamily, specifically the canonical TRPC family. Members of this family are activated through PLC-coupled receptors and function as receptor-regulated cation channels. The TRPC3/6/7 subfamily can be activated by DAG analogs, suggesting a mechanism of activation via PLC-coupled receptors.
When expressed in transfected cells, TRPC6 acts as a non-selective, store-independent, receptor-activated cation channel. It is activated by DAG in a PKC-independent manner and insensitive to IP3 activation.
TRPC6 is implicated in endogenous DAG-activated receptor-operated cation channels in vivo.
Diseases associated with TRPC6 include Glomerulosclerosis, Focal Segmental, 2 and Focal Segmental Glomerulosclerosis.
