Product Specific Information

Description:
The D7 monoclonal antibody reacts with mouse Sca-1, an 18 kDa member of the Ly-6 family of GPI-linked surface proteins. D7 recognizes both Ly-6E.1 and Ly-6A.2 molecules expressed by mouse hematopoietic stem cells, myeloid populations, and peripheral T and B cells. Sca-1 expression levels vary by strain and activation state, serving as a key marker for mouse hematopoietic progenitor/stem cell subsets.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.5 µg per test (100 µL final volume). Optimal dilution should be determined empirically.

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Use a 660/20 bandpass filter. Ensure the instrument can detect this fluorochrome.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored up to 3 days at 4°C in the dark using:

• IC Fixation Buffer (Product # 00-8222)
• 1-step Fix/Lyse Solution (Product # 00-5333)

Target Information

Stem Cell Antigen-1 (Sca-1):
Sca-1 is part of the Ly-6 family and is expressed on multipotent hematopoietic stem cells (HSC). It is widely used as a marker for murine HSC in combination with lineage depletion antibodies. Expression is found in fetal liver, adult bone marrow, peripheral blood, and spleen. Sca-1 may play a role in B and T cell activation regulation.
