Target Information

The Map kinase-interacting kinases (MKNK1 and MKNK2) are activated by Map kinases and regulate protein synthesis by phosphorylating eIF4E on Ser209, potentially contributing to tumorigenesis. Overexpression of Mnk2 increases phosphorylation of endogenous eIF-4E, indicating its role as an eIF-4E kinase in vivo. Mnk2 may also participate in cellular responses to environmental stress and cytokines. Active mutants of MNK1 and MNK2 reduce cap-dependent translation in 293 cells. Human Mnk2 shows ~94% identity with murine Mnk2 and 71% with human Mnk1. In vitro studies indicate Mnk2 is a better substrate than Mnk1 for Erk2, p38MAPKα, and p38MAPKβ.
