Background
This gene was identified as a tumor suppressor mutated in many cancers. The encoded protein is a phosphatidylinositol-3,4,5-trisphosphate 3-phosphatase with a tensin-like and catalytic domain similar to dual specificity protein tyrosine phosphatases. It preferentially dephosphorylates phosphoinositide substrates, negatively regulating intracellular phosphatidylinositol-3,4,5-trisphosphate levels and AKT/PKB signaling. A non-canonical upstream initiation site produces a longer isoform associated with the mitochondrial inner membrane, potentially involved in energy regulation.
