Background
The 26S proteasome is a multicatalytic proteinase complex with a highly ordered structure composed of two complexes: a 20S core and a 19S regulator.
The 20S core consists of four rings of 28 non-identical subunits (two rings of 7 alpha subunits and two rings of 7 beta subunits).
The 19S regulator includes a base with 6 ATPase subunits and 2 non-ATPase subunits, and a lid containing up to 10 non-ATPase subunits.
Proteasomes are abundant in eukaryotic cells and mediate ATP/ubiquitin-dependent peptide cleavage in a non-lysosomal pathway.
The PSMD3 gene encodes a non-ATPase subunit of the 19S regulator lid within the proteasome subunit S3 family.
Single nucleotide polymorphisms in this gene are associated with neutrophil count.
