Background
Alzheimer’s disease (AD) patients with inherited forms of the disease carry mutations in the presenilin proteins (PSEN1 or PSEN2) or the amyloid precursor protein (APP). These mutations increase the production of the longer form of amyloid-beta, the main component of amyloid deposits in AD brains. Presenilins are believed to regulate APP processing via gamma-secretase, an enzyme that cleaves APP. They may also be involved in Notch receptor cleavage, either by regulating gamma-secretase activity or acting as protease enzymes themselves. Two alternatively spliced transcript variants encoding different isoforms of PSEN2 have been identified.
