Background

Protein kinase C (PKC) is a family of serine- and threonine-specific protein kinases activated by calcium and diacylglycerol. PKC family members phosphorylate a wide range of protein targets and are involved in diverse cellular signaling pathways. They also act as major receptors for phorbol esters, a class of tumor promoters. Each PKC isoform has a distinct expression profile and role. PKC θ is a calcium-independent, phospholipid-dependent kinase crucial for T-cell activation, required for activation of NF-κB and AP-1 transcription factors, and may link TCR signaling to transcriptional activation.
