Product Specific Information

This F38-2E2 monoclonal antibody reacts with human CD366, also known as T cell immunoglobulin and mucin domain-containing protein 3 (TIM3) or HAVCR2.
CD366 is expressed on activated CD4+ T cell subsets (Th1, Th17, Treg), CD8+ T cells, monocytes, dendritic cells, and mast cells. It exists in membrane-bound and soluble forms, with Galectin-9 identified as its ligand. CD366 negatively regulates CD4+ T cells and influences Th1/Th17 cytokine secretion. It is implicated in tolerance, autoimmune diseases (e.g., multiple sclerosis), and HIV infection.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometry on stimulated human peripheral blood cells. Recommended use: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended detection filter: 660/20 bandpass. Ensure instrument compatibility.

Usage Recommendations:

  • When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific polymer interactions.
  • Protect from light to prevent photo-induced oxidation.
  • Fixed samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) at 4°C for up to 3 days.

Target Information

TIM3 (HAVCR2) is a 281-amino-acid, type I Th1-specific glycoprotein expressed on terminally differentiated CD4+ Th1 and CD8+ Tc1 cells.
It includes an IgV-like domain, a mucin-like extracellular domain, and a conserved tyrosine phosphorylation motif in the cytoplasmic region.
TIM3 is involved in macrophage activation, autoimmune disease induction, and immune tolerance regulation. It is associated with experimental autoimmune encephalomyelitis (EAE) and Th1-dependent autoimmune responses.
