• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

CD86 is one of two ligands (the other being CD80) for CTLA4 and CD28. It acts as a costimulatory molecule in eliciting T-cell help during antigen presentation.
In the absence of sufficient co-stimulation involving CD86/CD80, antigen presentation can induce tolerance.
CD86 is a type I membrane protein and a member of the immunoglobulin superfamily, expressed by antigen-presenting cells.
It binds to CD28 and CTLA4 on T cells:

• CD28 binding provides a costimulatory signal for T-cell activation.
• CTLA4 binding negatively regulates T-cell activation and diminishes immune response.

Alternative splicing results in two transcript variants encoding different isoforms of CD86. Additional variants have been described but not fully characterized.
Diseases associated with CD86 dysfunction include gallbladder squamous cell carcinoma and myocarditis.
