Target Information

p21-activated kinases (PAKs) are serine/threonine kinases involved in controlling various cellular processes such as cell cycle regulation, cytoskeleton dynamics, apoptosis, oncogenic transformation, and transcription. They contain a p21-binding domain and are regulated by small GTP-binding proteins Rac and Cdc42, and lipids that stimulate autophosphorylation and substrate phosphorylation.

PAK4 is activated by phosphorylation at Ser-474 and is implicated in oncogenesis, being overexpressed in multiple human tumor cell lines. It plays roles in cytoskeleton regulation, cell migration, proliferation, and survival by phosphorylating targets including RAF1, BAD, and CTNND1. PAK4 stabilizes microtubules via MARK2 inhibition and modulates actin networks, influencing cell morphology and adhesion.
