Target Information
Reversible phosphorylation of tyrosine residues is a key regulatory mechanism for numerous cellular events such as proliferation, differentiation, gene expression, and migration. Abnormalities in tyrosine phosphorylation contribute to various human diseases, including cancer and immune deficiencies.
There are at least 107 genes coding for protein tyrosine phosphatases (PTPs) in the human genome. All known PTPs possess a conserved 12 amino acid catalytic domain and differ by additional structural motifs.
Receptor-like PTPs have extracellular domains, a transmembrane region, and one or two intracellular phosphatase domains. Nonreceptor cytoplasmic PTPs contain a single phosphatase domain and additional motifs homologous to other protein classes.
PTPN5 (STEP), PTPRR, and PTPN7 form a family of phosphatases that specifically inactivate MAPKs (mitogen-activated protein kinases). Multiple STEP isoforms are found in the adult rat brain and are differentially enriched in regions associated with cognitive, affective, and motor behavior. Some isoforms arise through alternative splicing of the STEP gene, each with unique intracellular targets and functions.
