Product Description

Type I Collagen is the most abundant collagen type in the human body, serving as a major structural matrix protein in skin, bone, tendon, and fibrous connective tissues. It is a heterotrimer composed of one α2(I) chain and two α1(I) chains arranged in a triple helical conformation.

Collagen interacts with various cell surface receptors, especially integrins α1β1 and α2β1, facilitating cell attachment. HumaDerm exhibits all three molecular weight regions (α: 100 KDa, β: 200 KDa, γ: 300 KDa) with minimal fragmentation, making it ideal for coating culture surfaces or forming hydrogels.

Tissue Source

Type I collagen is isolated from human skin obtained from AATB-accredited and FDA-registered tissue banks and organ procurement organizations (OPOs). All tissues comply with 21 CFR 1271 requirements for transplantable human tissues.

Storage

Collagen-Coated Tissue Cultureware

Note: Optimize concentration and incubation conditions per cell type and application. Typical collagen concentration: 10–100 µg/ml.

1. Dilute HumaDerm in 10–20 mM HCl (pH 1.9–2.1) to 0.1 mg/ml.
2. Mix until homogeneous.
3. Aseptically coat tissue culture surfaces evenly.
4. Incubate at 37 °C for ≥ 2 hours.
5. Aspirate excess solution and air dry overnight.
6. Rinse with sterile PBS or medium.
7. Use immediately or store coated ware at 2–10 °C.

Making 3D Collagen Gels

Note: Perform all steps on ice or in a cold room (2–10 °C). HumaDerm forms gels at ≥ 0.5 mg/ml.

1. Dilute HumaDerm in 10–20 mM HCl (pH 1.9–2.1) to desired concentration.
2. Prepare chilled 10× medium or 10× PBS.
3. Mix 9 volumes collagen solution with 1 volume 10× buffer.
4. Gently swirl or pipette to mix.
5. Adjust pH to 7.0–8.0 (optimal ≈ 7.4).
6. Incubate at 37 °C for 30–120 minutes for gel formation.
