Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with the establishment and maintenance of pregnancy.

PR exists as two forms:

  • Form A (94 kDa)
  • Form B (120 kDa)

These forms arise from alternative translation initiation sites. PR-B is transcriptionally active and responsible for activating genes related to endometrial maintenance, pregnancy, and inhibition of ovulation. PR-A differs by a 165 amino acid deletion at the N-terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity.

In its inactive state, PgR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, conformational changes enable receptor dimerization and binding to progesterone response elements (PRE), resulting in activated transcription. Null mutations in the PGR gene lead to pleiotropic reproductive abnormalities.
