Target Information

Progression through the cell cycle requires activation of cyclin dependent kinases (Cdks). The monomeric catalytic subunit Cdk2 is inactive until partially activated by cyclin binding and fully activated by phosphorylation at Thr160. The enzyme responsible for this phosphorylation, known as Cdk-activating kinase (CAK), includes a catalytic subunit, a regulatory subunit, and a subunit of unknown function. The regulatory subunit, cyclin H, is necessary for Cdk7 activation. Mat1 (MNAT1) is a protein that associates with the cyclin H–Cdk7 complex throughout the cell cycle, forming an active Cdk-cyclin complex. Cyclin H–Cdk7–Mat1 complexes exhibit kinase activity toward Cdk activation domains and the C-terminal domain of RNA polymerase II, highlighting Mat1’s role as an essential assembly factor in Cdk-cyclin complex formation.
