Target Information

IL-21 is a 17 kDa immunomodulatory cytokine mainly produced by Natural Killer T (NKT) cells, T helper (Th)17, and T follicular helper (TFH) cells.
In TFH cells, IL-21 expression triggers autocrine signaling through IL-21R and STAT3, leading to transcriptional activation by Bcl6.
IL-21 is critical for TFH cell development and effector function, promoting T cell-dependent B cell differentiation, IgG production, and induction of apoptosis in naive B cells.
In Th17 cells, IL-21 enhances IL-23R expression and supports maturation and maintenance through STAT3, IRF4, and RORγt pathways while downregulating Foxp3.
IL-21 deficiency protects mice from chemically induced colitis, whereas elevated IL-21 levels are observed in colitis models.
