Target Information

The tumor suppressor protein p53 is a sequence-specific transcription factor activated by cellular stress. It mediates cell cycle arrest or apoptosis in response to DNA damage or nutrient deprivation. p53 regulates genes such as p21waf1, leading to G1 or G2/M arrest. Structurally, it includes an N-terminal transactivation domain, a central DNA-binding domain, an oligomerization domain, and a C-terminal regulatory domain. Phosphorylation at Ser15 is critical for activation and stabilization, while Ser392 phosphorylation is linked to tumor formation. p53 plays roles in cancer suppression, aging, and oxidative stress responses. It is typically expressed at low levels in normal cells but is elevated in transformed cells, contributing to malignancy. TP53 gene mutations are common in human cancers and hereditary cancer syndromes such as Li-Fraumeni syndrome.
