Product Specific Information
This antibody recognizes CD40, a type I transmembrane glycoprotein belonging to the TNFR superfamily. CD40 is broadly expressed on antigen-presenting cells (APCs) such as dendritic cells, B cells, macrophages, and monocytes, as well as non-immune endothelial and epithelial cells.
Upon binding to its ligand CD154, CD40 functions as a costimulatory molecule for activation of B cells, dendritic cells, monocytes, and other APCs.
Target Information
The CD40 antigen is a single-chain glycoprotein and a member of the tumor necrosis factor/nerve growth factor superfamily, showing homology to CD30.
CD40 is expressed on all B cells except plasma cells and is found on epithelial cells, carcinomas, and lymphoid dendritic cells. It plays a key role in immune and inflammatory responses, including:
- T cell-dependent immunoglobulin class switching
- Memory B cell development
- Germinal center formation
The AT-hook transcription factor AKNA regulates CD40 expression, facilitating cell interactions.
Adaptor protein TNFR2 interacts with CD40, mediating signal transduction.
The CD40-CD154 interaction is necessary for amyloid-beta-induced microglial activation, implicated in early Alzheimer’s disease pathogenesis.
Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
Diseases associated with CD40 dysfunction include Type 3 Hyper-IgM immunodeficiency and CD40 ligand deficiency.
For Research Use Only.
Not for use in diagnostic procedures.
