- Specificity Comments: Complement component proteins (C2, C3, C4, C5) act as potent anaphylatoxins released during complement activation. Their binding to G protein-coupled receptors (C3aR, C1R, C5aR) triggers proinflammatory responses such as cellular degranulation, smooth muscle contraction, arachidonic acid metabolism, cytokine release, leukocyte activation, and chemotaxis.
C3d, a terminal degradation product of C3, modulates adaptive immune responses via interaction with complement receptor type 2 (CR2). CR2 is crucial for high-affinity, memory humoral immune responses and natural antibody repertoire development. The pH- and ionic strength-dependent association of C3d with CR2 links innate and adaptive immunity.
Target Information
C3 is a key component of the complement system where classical and alternative activation pathways converge at the C3 activation step, splitting C3 into C3a and C3b. The molecular mass of C3 is 185 kDa, consisting of two chains (110 kDa and 75 kDa) joined by disulfide bonds.
