A recommended positive control tissue for this product includes epithelial cells (pancreas, colon, thyroid, etc.), though controls are not limited to these types.
The primary antibody is intended for laboratory professional use in detecting Cytokeratin 17 in formalin-fixed, paraffin-embedded tissues via manual qualitative immunohistochemistry (IHC). It is used after tumor diagnosis by conventional histopathology.
Type I Cytokeratin 17 (CK17) shows distinctive localization in human epithelial appendages, including hair follicles that undergo cyclical growth. CK17 is induced early after acute skin injury, along with CK6 and CK16. Predominant CK17 expression, frequent CK8 and CK19 expression, and minimal CK6/CK16 and CK1/CK10 expression characterize basal cell carcinomas (BCC), suggesting differentiation toward undifferentiated follicular epithelia, likely hair bulge cells.
Pretreatment of deparaffinized tissue with heat-induced or enzymatic epitope retrieval is recommended. IHC staining visualizes antigens through sequential application of primary and secondary antibodies, enzyme complexes, and chromogenic substrates. The reaction product is observed under a light microscope to aid differential diagnosis.
A positive tissue control must accompany every staining procedure. This control should include both positive and negative staining areas and confirm proper tissue preparation and staining. Weakly positive tissues are recommended for optimal quality control and reagent stability assessment.
Negative control tissues, internal or external, should confirm the absence of specific staining and help detect non-specific background staining. If specific staining occurs in negative controls, patient specimen results are invalid.
Target Information
Cytokeratins are intermediate filament proteins with biochemical diversity, represented by at least 20 human epithelial polypeptides (40–68 kDa, pI 4.9–7.8). Cytokeratin subtype expression patterns are characteristic of epithelial type and maturation stage. These patterns assist in distinguishing epithelial malignancies. Cytokeratin antibodies are valuable in immunohistochemistry, cytopathology, and flow cytometry for tumor differentiation.
