Target Information

AHR (Ah Receptor) belongs to a family of proteins including its dimerization partner ARNT (HIF-1 Beta) and Drosophila proteins PER and SIM.
AHR contains an N-terminal PAS domain (~200 amino acids) and is found in various tissues. It binds to specific DNA enhancer sequences and initiates transcription of the cytochrome P-450 (CYP1A1) gene.

AHR encodes a ligand-activated transcription factor involved in regulating biological responses to planar aromatic hydrocarbons. It regulates xenobiotic-metabolizing enzymes such as cytochrome P450, with ligands including polycyclic and halogenated aromatic compounds (e.g., TCDD).

Unlike steroid or thyroid hormone receptors, no known physiological ligand for AHR exists. In non-ligand activated cells, AHR complexes with HSP90 in the cytoplasm. Upon agonist binding, it transforms into a nuclear DNA-binding form, dissociates from HSP90, and forms a heterodimer with ARNT.

Diseases associated with AHR include eosinophilic fasciitis and seborrheic dermatitis.
