Target Information

CD30 (Ki-1, TNF Receptor Superfamily Member 8) is a type I transmembrane glycoprotein belonging to the TNF receptor superfamily.
Originally identified as a cell surface antigen on Hodgkin and Reed-Sternberg cells using monoclonal antibody Ki-1, CD30 interacts with its ligand CD30L (CD153) to mediate diverse biological effects including cell proliferation, activation, differentiation, and apoptosis.

CD30 plays a critical role in Hodgkin’s disease and other CD30+ lymphomas. It acts as a costimulatory molecule in thymic negative selection and is expressed on Hodgkin’s and Reed-Sternberg cells, some non-Hodgkin’s lymphomas, virus-infected T and B cells, and activated normal T/B cells.

In T cells, CD30 expression is associated with Th2-type cytokine-producing subsets and CD4+/CD8+ thymocytes co-expressing CD45RO and IL4 receptor. Soluble CD30 (sCD30) serves as a marker of Th2 immune response. TRAF2 and TRAF5 interact with CD30, activating NF-kappaB signaling.

CD30 is a positive regulator of apoptosis, limiting autoreactive CD8 effector T cell proliferation and protecting against autoimmunity. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.

CD30 is expressed by mononuclear cells in Hodgkin’s lymphoma, Reed-Sternberg cells, and most Anaplastic Large Cell Lymphomas (ALCL), as well as embryonal carcinomas.
