Product Specific Information

The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat Foxp3 proteins (FORKHEAD BOX P3, SCURFIN, JM2). Foxp3, a 49–55 kDa transcriptional regulator, is defective in scurfy mice. It is highly expressed in CD4⁺CD25⁺ regulatory T cells (Treg) and induces a Treg phenotype when expressed in CD4⁺CD25⁻ cells.

Epitope mapping shows reactivity to amino acids 75–125 of mouse Foxp3. Human Foxp3 shows alternative splicing in this region. Both isoforms are present in CD4⁺CD25⁺ lymphocytes. Mouse splenocytes lack this spliced isoform, suggesting species-specific regulation.

Applications Reported: Immunohistochemistry (frozen tissue, IHC-F) and Immunocytochemistry (ICC).
Applications Tested: Tested on frozen mouse spleen at ≤10 µg/mL. Not validated for flow cytometry.
Filter Recommendation: Use filter capturing 615 nm emission (e.g., Excitation 560/55, 585LP, Emission 645/75). Alexa Fluor® 594 filter acceptable.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis. It is highly expressed in CD25⁺CD4⁺ regulatory T cells, minimally in CD4⁺CD25⁻ cells, and absent in CD4⁻CD8⁺ T cells. FOXP3 mutations cause the scurfy phenotype in mice and IPEX syndrome in humans. Transcript variants encoding different isoforms exist, and FOXP3 expression in human cancer cells is regulated by p53 in response to DNA damage.
