세포 내 위치 (Cellular Localization)
Cytoplasm, nucleus, cell membrane (peripheral membrane protein), and cell junctions (focal adhesion).
Tyrosine phosphorylation occurs mainly at the cell membrane. Translocation to the membrane follows insulin stimulation via GRB14 and INSR binding. SRC and HSP90 promote membrane localization.
Nuclear localization depends on association with PTPN6 and phosphorylation at Ser-396.
In neuronal cells, restricted to nucleus; in non-neuronal cells, found in cytoplasm.
Ser-241 phosphorylated form localizes perinuclearly in neuronal cells and both nucleus and cytoplasm in non-neuronal cells.
IGF1 transiently increases phosphorylation at Ser-241 of neuronal PDPK1.
Background
- Catalytic activity: ATP + a protein → ADP + a phosphoprotein
- Function: Phosphorylates and activates PKB/AKT, PKA, PKC-zeta, RPS6KA1, and RPS6KB1. Plays a general role in signaling and development.
- PTM: Phosphorylated on tyrosine and serine/threonine residues. Ser-241 phosphorylation is required for full activity and can occur via autophosphorylation.
- Similarity: Belongs to the protein kinase superfamily, AGC Ser/Thr protein kinase family, PDK1 subfamily. Contains one PH domain and one protein kinase domain.
- Subcellular location: Membrane-associated after stimulation; tyrosine phosphorylation mainly at plasma membrane.
- Subunit interaction: Interacts with TUSC4.
- Tissue specificity: Expressed ubiquitously.
