세포 내 위치
Cytoplasm, nucleus, and cell membrane (peripheral membrane protein). Also found in cell junctions and focal adhesions. Tyrosine phosphorylation mainly occurs at the cell membrane. Translocates to the cell membrane following insulin stimulation via GRB14 and INSR binding. SRC and HSP90 assist in membrane localization. Nuclear localization depends on interaction with PTPN6 and phosphorylation at Ser-396. In neuronal cells, the Ser-241 phosphorylated form is perinuclear; in non-neuronal cells, it is found in both nucleus and cytoplasm. IGF1 transiently increases phosphorylation at Ser-241 in neuronal PDPK1.
Background
PDK1 is a serine/threonine kinase that phosphorylates and activates PKB/AKT, PKA, PKC-zeta, RPS6KA1, and RPS6KB1, playing a key role in signaling and development. Phosphorylation on Ser-241 is essential for full activity, and PDK1 can autophosphorylate this residue. It belongs to the AGC Ser/Thr protein kinase family, PDK1 subfamily, containing one PH domain and one protein kinase domain. PDK1 is membrane-associated after stimulation, interacts with TUSC4, and is ubiquitously expressed.
