Cytoplasm; Cell junction (focal adhesion); Cell projection (lamellipodium, ruffle membrane, invadopodium); Cell membrane; Nucleus (nucleoplasm, chromosome); Cytoskeleton (microtubule organizing center, centrosome).
Recruited to the cell membrane via interaction with CDC42 and RAC1, and to focal adhesions upon activation. Colocalized with CIB1 within membrane ruffles during cell spreading. Translocates to the nucleoplasm upon DNA damage when phosphorylated at Thr-212.
Background
This gene encodes a family member of serine/threonine p21-activating kinases (PAK proteins). These proteins are key effectors linking RhoGTPases to cytoskeleton reorganization and nuclear signaling. They regulate cell motility and morphology and serve as targets for small GTP binding proteins Cdc42 and Rac. Alternatively spliced transcript variants encoding different isoforms have been identified.
