Target Information
VEGF (vascular endothelial growth factor)는 45 kDa의 이황화 결합된 호모다이머 글라이코프로틴으로, 혈관신생(angiogenesis)에 관여하며 종양의 성장과 전이를 촉진합니다. VEGF는 내피세포의 생존, 유사분열, 화학주성 및 혈관 투과성 증가를 유도합니다.
VEGF 패밀리는 VEGF-A, VEGF-B, VEGF-C, VEGF-D, VEGF-E, 그리고 PIGF를 포함하며, VEGF와 그 수용체 시스템은 혈관신생 세포 신호의 핵심 조절자로 알려져 있습니다. 대부분의 종양은 혈관신생을 필요로 하며, VEGF는 이 과정과 관련된 가장 강력한 혈관신생 사이토카인으로 알려져 있습니다.
VEGF는 하나의 유전자에서 대체 스플라이싱을 통해 생성된 5가지 아이소폼을 가지며, 이들은 주로 내피세포에서 발현되는 두 가지 티로신 키나아제 수용체인 flt-1 (VEGFR-1)과 flk-1/KDR (VEGFR-2)에 결합합니다. 또한 FLT1의 대체 스플라이싱 형태인 sFLT1은 VEGF를 높은 친화도로 결합하여 혈관신생을 억제하는 역할을 합니다.
VEGF의 상승된 수준은 POEMS 증후군(Polyneuropathy, Organomegaly, Endocrinopathy, Monoclonal gammopathy, Skin changes)과 연관되어 있으며, 이는 여러 장기에 영향을 미칩니다.
