Target Information
PUMA는 Bax 및 Noxa를 포함하는 pro-apoptotic Bcl-2 가족 단백질 중 하나로, 모두 p53의 전사적 표적입니다. PUMA 유전자는 BH3 도메인을 포함한 두 단백질(PUMA-a 및 PUMA-b)을 암호화합니다. PUMA 단백질은 Bcl-2에 결합하고 미토콘드리아에 위치하며, p53 자극 시 cytochrome c 방출 및 세포자멸(apoptosis)을 유도합니다. PUMA는 p53 유도 세포자멸의 직접적인 매개체로 작용할 수 있으며, PUMA 발현 억제 시 p53에 의한 세포자멸 반응이 감소하는 것으로 알려져 있습니다. 따라서 PUMA는 cytochrome c/Apaf-1 의존 경로를 통한 p53 매개 세포사멸에 중요한 역할을 합니다.
